I had never used it Python.
However, I know deep learning.
To implement, do I have to use it?
Can use for it with matlab?

From my experience both (Matlab, or Octave) and Python can complement each other. Python is proven to be faster than Matlab, in particular some of the libraries developed for machine learning (such as TensorFlow) as these are largely written in C / C++.

Also, building some ML applications using your own coding helps tremendously in developing a much deeper understanding of deep learning and related concepts.

Hence in short, obviously you don't have to, but I strongly recommend it (learning Python). This portal offers a very good series of courses to get started ('Python for everybody').

It is worth the effort to learn Python. It will not be difficult to learn, especially if you already know Matlab. Spend a day or two learning about lists, tuples and dictionaries and then move right to numpy, pandas and matplotlib. You’ll know enough to be productive very quickly.
